The MTT Cell Viability Assay Kit provides a simple method for determining live cell numbers using a standard colorimetric plate readers. Determination of live cell numbers is often used to assess the rate of cell proliferation and cytotoxicity caused by drugs and cytotoxic agents. Among all non-radioactive viability assays, MTT assay developed by Mossman is one of the most versatile and popular assays. MTT is a tetrazolium salt that is turned into a purple formazan product after reduction by mitochondrial enzymes that are only present in metabolically active live cells, not in dead cells. The amount of formazan product generated is proportional to the number of living cells in the sample. The CytoSelect™ WST-1 Cell Proliferation Assay Reagent provides a colorimetric format for measuring and monitoring cell proliferation. Cells can be plated and then treated with compounds or agents that affect proliferation. Cells are then detected with the proliferation reagent, which is converted in live cells from WST-1 to the formazan form in the presence of cellular NADH and an electron mediator. An increase in cell proliferation is accompanied by increased signal, while a decrease in cell proliferation (and signal) can indicate the toxic effects of compounds or suboptimal culture conditions. The assay principles are basic and can be applied to most eukaryotic cell lines, including adherent and non-adherent cells and certain tissues. Water-soluble tetrazolium salts (WSTs) were developed by introducing positive or negative charges and hydroxy groups to the phenyl ring of the tetrazolium salt. Positive charges, such as trialkylammonio groups, improve the water-solubility of the formazan dye. However, a large cation is easily precipitated out with organic anions such as carboxylate or phosphate. Though a hydroxy group also improved the water-solubility of the tetrazolium salt, the corresponding formazan dye was not sufficiently water-soluble. Dojindo’s WSTs have sulfonate groups added directly or indirectly to the phenyl ring to improve water-solubility. Dojindo also offers several newly developed phenlyazo-type tetrazolium salts, which are easily reduced with NADH or other reducing agents to give orange or purple formazan dyes. Due to the phenylazo group, the color changes with heavy metal ions. Since the water solubility of Dojindo’s WSTs is high, 10 mM to 100 mM solutions can be prepared.
